  1. Glycosylated or deglycosylated MpXyn10A was incubated with Avicel or oat spelt xylan for 30 min at 4°C under agitation in 50 mM sodium acetate buffer, pH 5.5. The supernatant was separated from the polysaccharide, and the xylanolytic activity of each fraction was measured by incubation with 0.5% birchwood xylan at 70°C for 10 min. The amount of reducing sugars released was measured by the Miller method [33].
