Investigation of the binding specificity of the expansin dockerin with various cohesins. The specific binding of the CclEXL1 dockerin was determined by affinity-based ELISA. Microtitter plates of 96 wells were coated with CclEXL1, and (a) five CBM-Coh fusion proteins from C. clariflavum (Cohesins A1, A5, A8, D3, and G) or (b) eight CBM-Coh fusion proteins from different bacterial species (C. clariflavum, C. cellulolyticum, C. thermocellum, B. cellulosolvens, A. cellulolyticus, R. flavefaciens, and A. fulgidus) were added to the wells in increasing concentrations.
