Detection of mutations at the HCT_gRNA14 target site in T2 pNST3::CAS9-pU6::HCT_gRNA14 plants. Genomic DNA was isolated from leaf and stem tissues, and genomic sequences spanning the HCT_gRNA14 target site were PCR amplified and analyzed by MiSeq sequencing. Mutation frequencies at 4 bp before the PAM sequence at the target site are shown. Values are mean ± SD of two biological replicates
