Colorimetric assay and released acid determination. a The pH indicator-based colorimetric assay was performed to verify the activity of HaSGNH1 and its variants toward glyceryl tributyrate. Hydrolysis of glyceryl tributyrate was monitored by color changes from red to yellow due to a pH drop caused by released acid [31]. b The amount of acetic acid released from the hydrolysis of carbohydrate acetates by HaSGNH1 and its variants. Quantifications of released acetic acid from acetylated substrates were determined using an acetic acid kit following the manufacturer’s instructions (K-ACET, Megazyme, USA)
