In vivo validation of identified genes for stress tolerance. The relative fitness of E. coli harboring each gene involved in oxidative phosphorylation, electron transfer chain, efflux pump, and cell wall biogenesis under a ethanol (4%, v/v) and b HMF (20 mM) stresses. E. coli BL21(DE3) harboring each gene was incubated in 100 mL of LB broth at 37 °C for ethanol tolerance and was incubated in spectrophotometer (200 µL scale) for HMF tolerance. Finally, the relative fitness was calculated as described in “Methods”. The experimental data present means ± standard deviations from three independent experiments (**p-value < 0.01; ***p-value < 0.001)
