Released reducing ends and galactose monomers upon addition of a complex with multiple mannanase copies. All tested complexes contained Tf-Manna_Li-G_Doc-Ac (construct nr. 26), Tf-Manno_Doc-Bc (construct nr. 39) and Cj-Aga_Li-D_Doc-CtI (construct nr. 63) and one of the following scaffoldins: Ct-CBM3_Coh-Ac_Coh-Bc_Coh-CtI (construct nr. 135), Ct-CBM3_Coh-Ac_Coh-Ac_Coh-Bc_Coh-CtI (construct nr. 150), Ct-CBM3_Coh-Ac_Coh-Ac_Coh-Ac_Coh-Bc_Coh-CtI (construct nr. 151). Reaction mixtures were composed of 1 mL 0.5% GM and 10 pmol designer cellulosome. Enzymatic activity was determined quantitatively by measuring the released reducing sugars and galactose over time. An ANOVA followed by a post hoc Tukey test revealed a significant variation among the tested complexes
