The predicted and actual (measured) BPNPG5 activity of S. cerevisiae strains producing Cel3A and Cel5A. All 160 randomly selected strains containing in vitro SCRaMbLEd plasmids were cultivated for 48 h and the culture supernatants were used for enzyme assays; Cel3A activity was measured on pNPG, Cel5A activity was measured with DNS assay on CMC and the synergistic activity was measured on BPNPG5. Predicted synergistic activity was calculated using an equation generated by multiple linear regression analysis. Compared to the actual (measured) BPNPG5 activities, this model was highly effective in predicting BPNPG5 activity based on Cel3A and Cel5A (R2 = 0.97)
