Irreversible inactivation of Tr Cel7A is not responsible for the decreased hydrolysis rates at higher temperatures. 14C-BC (0.25 mg ml-1) was incubated with 0.25 μM TrCel7A, supplemented with 0.025 μM EG (TrCel5A) and 0.06 μM N188BG. Temperature was 40°C (◊) and 55°C (□). In one trial, the hydrolysis was conducted at 55°C for 30 min, and then the temperature was decreased (indicated by arrowhead) to 40°C (■).
